- Acentric chromosome fragmentation vector targeting Alu sequences. Contains promoters for in vitro RNA synthesis. [1] Constructed from pBP103 (ATCC 77087) by inserting a 0.3 kb Alu-containing sequence from BLUR8 into the BamHI site. pBP108 (ATCC 77088) and pBP109 (ATCC 77089) differ in the orientation of the Alu sequence. [1] Any unique restriction site between the targeting sequence and the telomere-adjacent sequence can be used to linearize the plasmid before transformation. [1] The EcoRV and ClaI sites between the SP6 promoter and the telomere sequence can be used for recovery from deletion derivatives of YAC insert sequences adjacent to the fragmentation site. [1] Restriction digests of the clone give the following sizes (kb): BamHI--4.6,0.3; EcoRI--4.9; SmaI--4.9; PstI--3.4, 1.15, 0.35.
