- This is a promoter cloning plasmid vector in which functional promoters are identified by conferring tetracycline resistance. [1] There are translation stop codons in all three reading frames between the EcoRI and HindIII cloning sites and the initiation codon for the tetracycline resistance gene. [1] This was constructed from pPV33 (ATCC 37073) by replacing the EcoRI site with two EcoRI sites flanking a HindIII site. [2] Medium is 1227 LB plus ampicillin. Hosts: E.coli RR1, E.coli.
