- The sequence reported here is a modified version of the type II methotrexate-resistant dihydrofolate reductase (DHFR) gene, R67 (see seperate entry). The 3' end of the original sequence was modified to determine if a conserved region of the gene encodes the active site of the protein. After modification, trimethoprim resistance in E. coli was observed at a comparable level to the one obtained with the unmodified gene, thus supporting the idea of a conserved region.
